Electrophoresis of Mucosal Homogenates

چکیده
The pepsinogens of human gastric mucosa have been the subject of several studies using electrophoretic and chromatographic techniques (Uriel, 1960; Seijffers, Segal, and Miller, 1963; Tang and Tang, 1963; Kushner, Rapp, and Burtin, 1964; Rapp, Aronson, Burtin, and Grabar, 1964; Hanley, Boyer, and Naughton, 1966; Taylor, 1968; Samloff, 1969); it has been shown that gel electrophoresis is superior to other fractionation procedures (Taylor, 1968) but the findings have been divergent. The present study reinvestigates this topic using the collective experience of other workers (Uriel, 1960; Seijffers et al, 1963; Tang and Tang, 1963; Kushner et al, 1964; Rapp et al, 1964; Hanley et al, 1966; Taylor, 1968; Samloff, 1969), defines the pepsinogen pattern of human gastric mucosa, explains the divergent findings ofearlier investigators, investigates the role of gastric disease in thedetermination ofthe pepsinogen pattern, describes a satisfactory column chromatographic method of pepsinogen fractionation, and relates the mucosal pepsinogen pattern to the pepsin patterns of human gastric juice.

Studies on the heterogeneity of human gastric zymogens.
THE PEPTIC ZYMOGENS OF HUMAN GASTRIC MUCOSA HAVE BEEN STUDIED IN THE PAST BY THREE DIFFERENT TECHNIQUES: phosphate-gradient chromatography on a column of DEAE-cellulose, chloride-gradient chromatography on DEAE-cellulose and agar-gel electrophoresis. These techniques have given different results, each suggesting the presence of a different number of pepsin zymogens in human gastric mucosa. In t...
متن کاملaB-Crystallin Selectively Targets Intermediate Filament Proteins during Thermal Stress
RESULTS. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and western immunoblot analyses showed selective interactions in lens cell homogenates between MBP-aB and endogenous aAand aB-crystallins, the lens-specific intermediate filament proteins phakinin (CP49) and filensin (CP115), and vimentin during a mild 20-minute heat shock at 45°C. No interactions were observed with t...
متن کاملHistochemical and biochemical analysis of the size-dependent nanoimmunoresponse in mouse Peyer’s patches using fluorescent organosilica particles
BACKGROUND/OBJECTIVE The size-dependent mucosal immunoresponse against nanomaterials (nanoimmunoresponse) is an important approach for mucosal vaccination. In the present work, the size-dependent nanoimmunoresponse of mouse Peyer's patches (PPs) and immunoglobulin A (IgA) level was investigated using fluorescent thiol-organosilica particles. METHODS Various sizes of fluorescent thiol-organosi...
متن کاملStarch-gel electrophoresis of the soluble lens proteins from normal and galactosemic animals.
The soluble proteins of normal calf, rabbit, and rat lenses have been studied by the method of starch-gel electrophoresis. The electrophoretic separations of lens homogenates are compared to those of alpha, beta, and gamma crystallins prepared by paper electrophoresis and by precipitation and gel filtration techniques. Rabbit lens solutions are separated into a total of 15 protein zones, rat le...
متن کاملAlphaB-crystallin selectively targets intermediate filament proteins during thermal stress.
PURPOSE AlphaB-Crystallin is a small heat shock protein (sHsp) expressed at high levels in the lens of the eye, where its molecular chaperone functions may protect against cataract formation in vivo. The purpose of this study was to identify protein targets for the sHsp alphaB-crystallin in lens cell homogenates during conditions of mild thermal stress. METHODS The authors report the use of a...
